Abstract

ANTIBACTERIAL ACTIVITY TEST OF JAVA CHILI LEAVES EXTRACT (Piper retrofractum vahl) ON Eschericia coli GROWTH ABSTRACT Diarrhea is caused by infection with the bacterium Escherichia coli, which is transmitted through the fecal-oral route. The use of natural materials can serve as an alternative treatment for various bacterial infections, including the leaves of the Javanese chili pepper (Piper retrofractum Vahl). This study aimed to investigate the antibacterial activity of Javanese chili pepper leaf extract against the growth of Escherichia coli at concentrations of 25%, 50%, 75%, and 100%. The research was conducted as a Quasi Experimental study using a Static Group Comparison Design and the Kirby-Bauer disk diffusion method. Each concentration was tested with five repetitions. Statistical tests included the Shapiro-Wilk test and the Kruskal-Wallis test. Data analysis was performed using the Kruskal-Wallis Multiple Comparisons test. The tested extract concentrations were 25%, 50%, 75%, and 100%, with Ciprofloxacin used as the positive control. Antibacterial activity was indicated by the formation of inhibition zones at 50%, 75%, and 100% extract concentrations, with average diameters of 7.39 mm, 7.80 mm, and 9.61 mm, respectively. No inhibition zone was observed at 25%. The antibacterial effect at 50%, 75%, and 100% was classified as moderate. Among the concentrations tested, a significant difference was found only between the 50% extract concentration and the positive control, with a p-value of 0.003. UJI AKTIVITAS ANTIBAKTERI EKSTRAK DAUN CABAI JAWA (Piper retrofractum vahl)TERHADAP PERTUMBUHAN BAKTERI Eschericia coli ABSTRAK Diare disebabkan oleh infeksi bakteri Escherichia coli yang ditularkan secara fecal oral. Pemanfaatan bahan alam dapat menjadi alternatif pengobatan berbagai jenis penyakit infeksi bakteri salah satunya daun cabai Jawa. Tujuan dari penelitian ini dilakukan untuk mengetahui aktivitas antibakteri daun cabai Jawa (Piper retrofractum vahl) terhadap pertumbuhan bakteri Escherichia coli pada konsentrasi 25%, 50%, 75%, dan 100%. Penelitian dilakukan secara Quasi Eksperimental dengan rancangan Static Group Comparison Disgn menggunakan metode difusi cakram cara Kirby-Bauer dengan masing-masing variasi konsentrasi dilakukan pengulangan sebanyak lima kali. Uji statistic yang digunakan adalah uji Shapiro Wilk dan uji Kruskal-Wallis Analisis data dilakukan dengan melakukan uji Kruskal-Wallis Multiple Comparions.Variasi konsentrasi ekstrak yang diuji yaitu konsentrasi 25%, 50%, 75%, 100% sebagai sampel uji dan antibiotik Ciprofloxacin sebagai kontrol kerja. Zona daya hambat antibakteri terhadap bakteri Escherichia coli yang ditunjukkan dengan terbentuknya zona hambat pada konsentrasi ekstrak 50%, 75% dan 100% dengan rerata diameter zona hambat masing-masing sebesar 7.39 mm; 7.80 mm; dan 9.61 mm. Sedangkan pada konsentrasi ekstrak 25% tidak terbentuk zona hambat. Kategori daya hambat antibakteri yang dimiliki pada konsentrasi 50%, 75%, dan 100% tergolong kedalam daya hambat sedang. Diantara variasi konsentrasi, hanya terdapat perbedaan signifikan pada konsentrasi ekstrak konsentrasi 50% dengan kontrol positif sebesar 0.003. Kata kunci : Ekstrak daun cabai Jawa; zona hambat
