Abstract
ANALYSIS OF PURITY AND CONCENTRATION OF Staphylococcus aureus DNA ISOLATED BY BOILING METHOD WITH MODIFIED ETHANOL PRECIPITATION ABSTRACT Background Pneumonia is an infectious disease caused by Staphylococcus aureus. Bacterial identification currently performed molecularly using PCR, which requires DNA isolation. The method of DNA isolation by cell boiling can be used. However, this method still requires modification in the form of adding an ethanol precipitation step. Aim of the study was to determine the results of the purity and concentration analysis of Staphylococcus aureus DNA isolated using the cell boiling method with ethanol precipitation modification. Method employed an experimental Posttest Only Control Group design using simple random sampling. Data were analyzed using the Independent Samples T-Test. Result qualitative test yielded 32 samples (100%) positive for detection of 16S rRNA gene. The quantitative test for cell boiling isolation without ethanol precipitation modification yielded an average concentration 61.4 ng/µl and a purity 2.24. For cell boiling isolation with ethanol precipitation modification, the average concentration 10.4 ng/µl and purity 1.32. Significant difference was found between the treatment groups, as evidenced by the results of the Independent Samples T-Test with a significant p-value 0.000. Conclusion The cell boiling isolation method without ethanol precipitation modification resulted better DNA purity and concentration compared to the isolation method with ethanol precipitation modification. Keyword: DNA purity, DNA concentration ANALISIS KEMURNIAN DAN KONSENTRASI DNA Staphylococcus aureus HASIL ISOLASI METODE PEREBUSAN DENGAN MODIFIKASI PRESIPITASI ETANOL ABSTRAK Latar belakang Pneumonia merupakan penyakit infeksi yang disebabkan oleh Staphylococcus aureus. Identifikasi bakteri kini dilakukan secara molekuler menggunakan PCR yang memerlukan isolasi DNA. Metode isolasi DNA dengan perebusan sel dapat digunakan, namun metode ini masih membutuhkan modifikasi berupa penambahan tahap presipitasi etanol. Tujuan penelitian untuk mengetahui hasil analisis kemurnian dan konsentrasi DNA Staphylococcus aureus hasil isolasi metode perebusan dengan modifikasi presipitasi etanol. Metode penelitian eksperimen desain Posttest Only Control Group dengan menggunakan simple random sampling. Data dianalisis dengan uji Independent Sample T-Test. Hasil uji kualitatif didapatkan 32 sampel (100%) positif terdeteksi gen 16S rRNA. Uji kuantitatif pada isolasi perebusan tanpa modifikasi presipitasi etanol didapatkan rata-rata konsentrasi 61,4 ng/µl dan kemurnian 2,24. Pada isolasi perebusan dengan modifikasi presipitasi etanol didapatkan rata-rata konsentrasi 10,4 ng/µl dan kemurnian 1,32. Terdapat perbedaan bermakna antara kelompok perlakuan yang dibuktikan melalui hasil uji statistik Independent Sample T-Test dengan dapatkan nilai signifikan 0,000. Simpulan metode isolasi perebusan tanpa modifikasi presipitasi etanol menghasilkan kemurnian dan konsentrasi DNA lebih baik dibandingkan metode isolasi dengan modifikasi presipitasi etanol.
