Abstract

THE EFFECT OF DELAY TIME ON EDTA PLASMA SAMPLES ON PROTHROMBIN TIME (PT) TEST RESULTS ABSTRACT Background: The Prothrombin Time (PT) test is an important parameter in assessing blood coagulation function, particularly the extrinsic pathway and the common pathway. PT test results are significantly influenced by preanalytical factors, such as the type of anticoagulant used and the delay in testing. The use of EDTA, which is not a standard anticoagulant for coagulation testing, as well as testing delays, has the potential to affect the stability of blood clotting factors. Objective: This study aims to determine the effect of testing delay on EDTA plasma samples on Prothrombin Time (PT) test results. Methods: This study used a true experimental design with a posttest-only control group approach. The study sample consisted of 9 participants with a total of 27 treatments, namely tests at 0 hours, 2 hours, and 4 hours. PT testing was performed using Sysmex CA-600. Results: The results showed that the delay time had a significant effect on the PT value of 0.025 (p < 0.05), where the longer the delay in testing, the greater the change in PT values, which tended to increase due to a decrease in coagulation factor stability. Conclusion: There is an effect of the delay time of the test on EDTA plasma samples on the PT test results. A longer delay can cause changes in PT values, which may potentially affect the interpretation of test results. Keywords: Prothrombin Time, EDTA, coagulation PENGARUH WAKTU PENUNDAAN PADA SAMPEL PLASMA EDTA TERHADAP HASIL PEMERIKSAAN PROTHROMBIN TIME (PT) ABSTRAK Latar Belakang: Pemeriksaan Prothrombin Time (PT) merupakan parameter penting dalam menilai fungsi koagulasi darah, khususnya jalur ekstrinsik dan jalur bersama. Hasil pemeriksaan PT sangat dipengaruhi oleh faktor pra-analitik, seperti jenis antikoagulan yang digunakan dan waktu penundaan pemeriksaan. Penggunaan EDTA yang bukan merupakan antikoagulan standar untuk pemeriksaan koagulasi, serta adanya penundaan pemeriksaan, berpotensi memengaruhi stabilitas faktor pembekuan darah. Tujuan: Penelitian ini bertujuan untuk mengetahui pengaruh waktu penundaan pemeriksaan pada sampel plasma EDTA terhadap hasil pemeriksaan Prothrombin Time (PT). Metode: Penelitian ini menggunakan desain true experimental dengan pendekatan posttest-only control group. Sampel penelitian terdiri dari 9 partisipan dengan total 27 perlakuan, yaitu pemeriksaan pada waktu Segera, 2 jam, dan 4 jam. Pemeriksaan PT dilakukan menggunakan alat Sysmex CA-600. Hasil: Hasil penelitian menunjukkan adanya pengaruh waktu penundaan terhadap nilai PT sebesar 0,025 (p < 0,05), di mana semakin lama waktu penundaan pemeriksaan, terjadi perubahan nilai PT yang cenderung meningkat akibat penurunan stabilitas faktor koagulasi. Simpulan: Terdapat pengaruh waktu penundaan pemeriksaan pada sampel plasma EDTA terhadap hasil pemeriksaan PT. Penundaan yang lebih lama dapat menyebabkan perubahan nilai PT sehingga berpotensi memengaruhi interpretasi hasil pemeriksaan. Kata Kunci: Prothrombin Time, EDTA, koagulasi
