Abstract

MOLECULAR IDENTIFICATION AND ANTIBACTERIAL ACTIVITY TEST OF LACTIC ACID BACTERIA ISOLATES FROM TEMPE LIQUID WASTE IN PADANGSAMBIAN KAJA ABSTRACT Increasing antibiotic resistance in pathogenic bacteria has encouraged the development of alternative natural antibacterial agents. Lactic acid bacteria (LAB) derived from tempeh wastewater have potential as antibacterial agents. This study aimed to perform molecular identification and evaluate the antibacterial activity of LAB isolates from tempeh liquid waste in Padangsambian Kaja. Methods included isolation on MRS Agar, phenotypic identification (catalase test, Gram staining, endospore staining, and fermentation type), and molecular identification using Polymerase Chain Reaction with 16S rRNA primers and Lactobacillus-specific primers. Antibacterial activity was tested using the agar well diffusion method against Staphylococcus aureus. The results showed that four isolates were obtained, with three isolates (A1, A2, and B) meeting LAB characteristics (Gram-positive, catalase-negative, non-spore-forming, and heterofermentative). PCR results revealed DNA bands of approximately 1.500 bp (16S rRNA) and 300-400 bp (Lactobacillus). Antibacterial activity showed inhibition zones of 11.18 mm, 11.54 mm, and 11.32 mm, categorized as strong. In conclusion, tempeh wastewater has potential as a source of LAB belonging to the Lactobacillus genus with antibacterial activity against Staphylococcus aureus. IDENTIFIKASI MOLEKULER DAN UJI AKTIVITAS ANTIBAKTERI ISOLAT BAKTERI ASAM LAKTAT DARI LIMBAH CAIR TEMPE DI PADANGSAMBIAN KAJA ABSTRAK Meningkatnya resistensi antibiotik pada bakteri patogen mendorong pengembangan alternatif antibakteri alami. Bakteri asam laktat (BAL) dari limbah cair tempe berpotensi sebagai agen antibakteri. Penelitian ini bertujuan untuk mengidentifikasi secara molekuler dan menguji aktivitas antibakteri isolat BAL dari limbah cair tempe di Padangsambian Kaja. Metode meliputi isolasi pada media MRS Agar, identifikasi fenotipik (katalase, Gram, endospora, fermentasi), serta identifikasi molekuler menggunakan metode Polymerase Chain Reaction dengan primer 16S rRNA dan primer spesifik Lactobacillus. Uji antibakteri dilakukan dengan metode difusi sumuran terhadap Staphylococcus aureus. Hasil menunjukkan diperoleh empat isolat, dengan tiga isolat (A1, A2, B) memenuhi karakteristik BAL (Gram positif, katalase negatif, non-spora, heterofermentatif). Hasil PCR menunjukkan pita DNA ±1.500 bp (16S rRNA) dan 300-400 bp (Lactobacillus). Uji antibakteri menunjukkan zona hambat berturut-turut sebesar 11,18 mm; 11,54 mm; dan 11,32 mm yang termasuk kategori kuat. Simpulan penelitian ini adalah limbah cair tempe berpotensi sebagai sumber BAL genus Lactobacillus yang memiliki aktivitas antibakteri terhadap Staphylococcus aureus.
