Abstract

ANTIBACTERIAL ACTIVITY TEST OF LACTIC ACID BACTERIA ISOLATES FROM TOFU WASTEWATER AGAINST Staphylococcus aureus ABSTRACT Staphylococcus aureus is a pathogenic bacterium that causes skin infections and has been reported to exhibit resistance. Lactic acid bacteria, which can be isolated from tofu wastewater, have the potential to act as natural antimicrobial agents through the production of bacteriocins, organic acids, hydrogen peroxide, and carbon dioxide. This study aims to assess the antibacterial activity of lactic acid bacteria isolates from tofu wastewater against Staphylococcus aureus. This research is a descriptive analytic study using a purposive sampling technique. Tofu wastewater samples were collected from 20 tofu factories in Klungkung District. Lactic acid bacteria were isolated on MRSA media and identified through catalase tests, Gram staining, and fermentation type tests, followed by antibacterial activity testing against Staphylococcus aureus using the well diffusion method. The results showed 22 isolates of lactic acid bacteria with characteristics of catalase-negative, Gram-positive, bacilli and cocci shapes, and homofermentative. All isolates exhibited antibacterial activity against Staphylococcus aureus, with an average inhibition zone diameter of 8.97 mm, categorized as moderate to strong. The conclusion of this study is that lactic acid bacteria isolates from tofu wastewater have antibacterial activity against Staphylococcus aureus. UJI AKTIVITAS ANTIBAKTERI ISOLAT BAKTERI ASAM LAKTAT DARI LIMBAH CAIR TAHU TERHADAP Staphylococcus aureus ABSTRAK Staphylococcus aureus merupakan bakteri patogen penyebab infeksi kulit yang dilaporkan mengalami resistensi. Bakteri asam laktat yang dapat diisolasi dari limbah cair tahu memiliki potensi sebagai agen antimikroba alami melalui produksi bakteriosin, asam organik, hidrogen perioksida, dan karbon dioksida. Penelitian ini bertujuan untuk mengetahui aktivitas antibakteri isolat bakteri asam laktat dari limbah cair tahu terhadap Staphylococcus aureus. Penelitian ini merupakan penelitian deskriptif analitik dengan menggunakan teknik purposive sampling. Sampel limbah cair tahu diambil sebanyak 20 sampel dari pabrik tahu di Kecamatan Klungkung. Isolasi bakteri asam laktat dilakukan pada media MRSA kemudian diidentifikasi melalui uji katalase, pewarnaan Gram, dan uji tipe fermentasi serta dilanjutkan dengan pengujian aktivitas antibakteri terhadap Staphylococcus aureus menggunakan metode sumuran. Hasil penelitian menunjukkan 22 isolat bakteri asam laktat dengan karakteristik katalase negatif, Gram positif, berbentuk basil dan kokus, serta bersifat homofermentatif. Seluruh isolat menunjukkan aktivitas antibakteri terhadap Staphylococcus aureus dengan rata-rata diameter zona hambat yaitu 8,97 mm, dikategorikan sebagai sedang hingga kuat. Simpulan penelitian ini adalah terdapat aktivitas antibakteri isolat bakteri asam laktat dari limbah cair tahu terhadap Staphylococcus aureus.
