Abstract

COMPARISON OF THE ANTIBACTERIAL EFFECTIVENESS OF Citrus juice (Citrus aurantifolia) AND HAND SANITIZER AGAINST THE GROWTH OF THE BACTERI Staphylococcus aureus ABSTRAC Lime juice (Citrus aurantifolia) contains active compounds such as flavonoids and citric acid, which are known to possess antibacterial properties. This study aimed to measure and compare the inhibitory zone effectiveness of lime juice and hand sanitizer at various concentrations (45%, 60%, and 75%) against the growth of Staphylococcus aureus ATCC 29213. The disc diffusion method was employed with four replications for each sample. The results showed that lime juice was able to form inhibitory zones with average diameters of 0.84 mm (45%), 1.66 mm (60%), and 3.75 mm (75%), respectively, while hand sanitizer produced no inhibitory zones at all tested concentrations (0 mm). The positive control using chloramphenicol antibiotic exhibited an inhibitory zone of 24.5 mm, whereas the negative control showed no inhibition. It can be concluded that lime juice possesses antibacterial activity against Staphylococcus aureus, with its effectiveness increasing along with concentration. However, its inhibitory effect was significantly lower than that of the antibiotic control. In contrast, hand sanitizer did not demonstrate antibacterial activity in this diffusion method, likely due to the high volatility of alcohol, which hindered its diffusion into the agar medium. PERBANDINGAN EFEKTIVITAS ANTIBAKTERI AIR PERASAN JERUK NIPIS (Citrus aurantifolia) DAN HAND SANITIZER TERHADAP PERTUMBUHAN BAKTERI Staphylococcus aureus ABSTRAK Air perasan jeruk nipis (Citrus aurantifolia) diketahui memiliki kandungan senyawa aktif seperti flavonoid dan asam sitrat yang berpotensi sebagai antibakteri. Penelitian ini bertujuan untuk mengukur dan membandingkan efektivitas zona hambat air perasan jeruk nipis dan hand sanitizer pada berbagai konsentrasi (45%, 60%, dan 75%) terhadap pertumbuhan bakteri Staphylococcus aureus ATCC 29213. Metode yang digunakan adalah uji difusi cakram. Hasil penelitian menunjukkan bahwa air perasan jeruk nipis mampu membentuk zona hambat dengan rata-rata diameter berturut-turut sebesar 0,84 mm (45%), 1,66 mm (60%), dan 3,75 mm (75%), sedangkan hand sanitizer tidak membentuk zona hambat pada semua konsentrasi yang diuji (0 mm). Kontrol positif menggunakan antibiotik kloramfenikol menghasilkan zona hambat sebesar 24,5 mm, sementara kontrol negatif tidak menunjukkan zona hambat. Dapat disimpulkan bahwa air perasan jeruk nipis memiliki aktivitas antibakteri terhadap Staphylococcus aureus, yang meningkat seiring bertambahnya konsentrasi, meskipun efektivitasnya masih jauh lebih rendah dibandingkan antibiotik. Sebaliknya, hand sanitizer tidak menunjukkan efektivitas antibakteri dalam metode uji ini, kemungkinan disebabkan oleh sifat volatilitas alkohol yang menghambat difusi ke media agar.
