Abstract
PERBEDAAN JENIS PELARUT TERHADAP KADAR TOTAL FENOL DAN AKTIVITAS ANTIOKSIDAN DARI EKSTRAK DAUN CENGKEH SECARA SPEKTROFOTOMETRI UV-VIS ABSTRAK Latar Belakang: Cengkeh merupakan salah satu sumber alami senyawa fenolik yang tinggi yang mempunyai aktivitas antioksidan yang dapat menangkal radikal bebas. Komposisi senyawa yang aktif dalam ekstrak daun cengkeh sangat bergantung pada kemampuan pelarut untuk mengekstraksi metabolit sekunder selama proses ekstraksi. Tujuan: Mengetahui perbedaan jenis pelarut terhadap kadar total fenol dan aktivitas antioksidan serta hubungan kadar total fenol dengan aktivitas antioksidan dari ekstrak daun cengkeh. Metode: Pengujian kadar total fenol dilakukan dengan metode Folin-Ciocalteu dan aktivitas antioksidan dengan metode DPPH (2,2-diphenyl-1-picryl-hydrazyl) serta uji parametrik One Way ANOVA untuk mengevaluasi perbedaan dari tiga jenis pelarut, yaitu etanol 96%, akuades, dan etil asetat terhadap kadar total fenol dan aktivitas antioksidan dalam ekstrak daun cengkeh. Hasil: Penelitian menunjukkan bahwa ekstrak akuades memiliki kandungan total fenol paling besar yaitu 668,54 mgGAE/g dan IC50 paling rendah yaitu 49,96 μg/ml, diikuti dengan total fenol ekstrak etanol 96% sebesar 634,09 mgGAE/g dan IC50 sebesar 70,29 μg/ml, sedangkan etil asetat memiliki kandungan total fenol paling rendah yaitu 600,84 mgGAE/g dan nilai IC50 yang paling tinggi 87,25 μg/ml. Kesimpulan: Terdapat perbedaan dan hubungan yang signifikan (p < 0,05) pada kadar total fenol dan aktivitas antioksidan dari ekstrak daun cengkeh menggunakan jenis pelarut yang berbeda. DIFFERENCE OF SOLVENT TYPES ON TOTAL PHENOL CONTENT AND ANTIOXIDANT ACTIVITY OF CLOVE LEAF EXTRACTS BY UV-VIS SPECTROPHOTOMETRY ABSTRACT Background: Clove is one of the natural sources of high phenolic compounds with antioxidant activity that can counteract free radicals. The composition of active compounds in clove leaf extracts highly depends on the solvent's ability to extract secondary metabolites during the extraction process. Objective: To determine the differences in solvent types on total phenol content and antioxidant activity, as well as the correlation between total phenol content and antioxidant activity of clove leaf extracts. Method: Total phenol content was tested using the Folin-Ciocalteu method and antioxidant activity using the DPPH (2,2-diphenyl-1-picryl-hydrazyl) method, along with the One-Way ANOVA parametric test to evaluate the differences among three types of solvents: 96% ethanol, distilled water, and ethyl acetate on total phenol content and antioxidant activity in clove leaf extracts. Results: The study showed that distilled water extract had the highest total phenol content at 668.54 mgGAE/g and the lowest IC50 at 49.96 μg/ml, followed by 96% ethanol extract with a total phenol content of 634.09 mgGAE/g and IC50 of 70.29 μg/ml, while ethyl acetate had the lowest total phenol content at 600.84 mgGAE/g and the highest IC50 value at 87.25 μg/ml. Conclusion: There were significant differences and correlations (p < 0.05) found between total phenol content and antioxidant activity.
