Abstract

DIFFERENCES IN HEMOGLOBIN LEVELS IN PACKED RED CELL COMPONENTS BEFORE AND AFTER BEING STORED FOR ONE WEEK IN THE BLOOD REFRIGERATOR OF THE PMI BLOOD DONOR UNIT IN DENPASAR CITY ABSTRACT Hemoglobin level in Packed Red Cell (PRC) component is an important parameter in every blood bag that will be transfused because it can improve the condition of anemia using blood that has good quality and has normal hemoglobin levels. Long storage of blood can cause many red blood cells to be damaged and fewer red blood cells to survive. Due to the large number of lysed red blood cells, blood stored for a long period of time will experience a change in the oxygen dissociation curve, which shifts to the left, so that oxygen is more strongly bound to hemoglobin and the amount of oxygen delivered to the tissue decreases. The purpose of this study was to determine the difference in hemoglobin levels in packed red cell components before and after being stored for one week in the blood refrigerator of the PMI Blood Donor Unit in Denpasar City. This type of research is Pre-Experimental with the form of One-Group-Pretest-Posttest Design with the number of samples used as many as 15 samples with purposive nonprobability sampling technique. Data collection through the results of checking hemoglobin levels using the POCT (Point Of Care Testing) tool. Based on the results of the Paired-T Test test, the p-value = 0.000 (p <0.05) which means that there is a significant difference between hemoglobin levels. Keywords: Hemoglobin, Storage Duration, PRC PERBEDAAN KADAR HEMOGLOBIN PADA KOMPONEN PACKED RED CELL SEBELUM DAN SESUDAH DISIMPAN SATU MINGGU DI BLOOD REFRIGERATOR UNIT DONOR DARAH PMI KOTA DENPASAR ABSTRAK Kadar hemoglobin pada komponen Packed Red Cell (PRC) adalah parameter penting pada setiap kantong darah yang akan ditransfusikan karena dapat dapat menyebabkan banyaknya sel darah merah yang rusak dan semakin sedikit sel darah merah yang bertahan hidup. Karena banyaknya sel darah merah yang lisis, darah yang disimpan dalam jangka waktu yang lama akan mengalami perubahan dalam kurvadisosiasi oksigen, yang bergeser ke arah kiri, sehingga oksigen lebih kuat terikat dengan hemoglobin dan jumlah oksigen yang diberikan ke jaringan berkurang. Karena banyaknya sel darah merah yang lisis maka kemungkinan darah yang disimpan akan mengalami kenaikkan kadar hemoglobin. Tujuan penelitian ini yaitu untuk mengetahui perbedaan kadar hemoglobin pada komponen packed red cell sebelum dan sesudah disimpan satu minggu di blood refrigerator Unit Donor Darah PMI Kota Denpasar. Jenis penelitian ini adalah Pre-Experimental dengan bentuk One-Group-Pretest-Posttest Design dengan jumlah sampel yang digunakan sebanyak 15 sampel dengan Teknik pengambilan sampel nonprobability sampling secara purposive. Pengumpulan data melalui hasil pemeriksaan kadar hemoglobin menggunakan alat POCT (Point Of Care Testing). Berdasarkan hasil uji Paired-T Test yang telah didapatkan hasil nilai p-value = 0,000 (p<0,05) yang artinya ada perbedaan yang signifikan antara kadar hemoglobin sebelum disimpan satu minggu di blood refrigerator dan kadar hemoglobin sesudah disimpan satu minggu di blood refrigerator..
