Abstract

ANTIBACTERIAL EFFECTIVENESS TEST OF SINGLE AND COMBINATION EXTRACTS OF PAPAYA AND GUAVA LEAVES ON THE GROWTH OF Escherichia Coli BACTERIA ABSTRACT Background: Escherichia coli is one of the pathogenic bacteria that cause diarrhea in humans. Papaya leaves and guava leaves contain antibacterial compounds in the form of tannins, alkaloids, flavonoids, saponins, and steroids. Purpose: This study aims to determine the effectiveness of papaya leaf and guava leaf extracts both singly and in combination against the growth of Escherichia coli bacteria. Methods: The experimental design used Quasi Experiment with Post-test Only Controlled Group Design of the well diffusion method with 5 treatments, namely papaya leaf extract, guava leaf extract, a combination of extracts with a ratio of 1: 1, 2: 1, and 1: 2. Results: papaya leaf extract and guava leaf extract both singly and in combination are able to inhibit the growth of Escherichia coli bacteria with the diameter of the inhibition zone of papaya leaf extract (11.21 mm), guava leaf extract (10.69 mm), 1:1 (6.95 mm), 2:1 (7.10 mm) and 1:2 (11.65 mm). Conclusion: The inhibition zone diameter results were categorized as moderate to strong. The results of the antibacterial effectiveness test by papaya leaf extract amounted to 32.63%, guava leaf extract amounted to 31.12%, 1:1 amounted to 20.23%, 2:1 amounted to 22.47%, and 1:2 amounted to 33.91%. It can be concluded that there are differences in antibacterial effectiveness between single and combined extracts of papaya leaves and guava leaves against the growth of Escherichia coli bacteria. Key words: Papaya leaf, guava leaf, Escherichia coli UJI EFEKTIVITAS ANTIBAKTERI EKSTRAK TUNGGAL DAN KOMBINASI DAUN PEPAYA DAN DAUN JAMBU BIJI TERHADAP PERTUMBUHAN BAKTERI Escherichia coli ABSTRAK Latar belakang: Escherichia coli merupakan salah satu bakteri patogen penyebab diare pada manusia. Daun pepaya dan daun jambu biji mengandung senyawa antibakteri berupa tanin, alkaloid, flavonoid, saponin, dan steroid. Tujuan: Penelitian ini bertujuan untuk mengetahui efektivitas ekstrak daun pepaya dan daun jambu biji baik tunggal maupun kombinasi terhadap pertumbuhan bakteri Escherichia coli. Metode: Rancangan eksperimen menggunakan Quasi Experiment dengan desain Post-test Only Controlled Group Design metode difusi sumuran dengan 5 perlakuan yaitu ekstrak daun pepaya, ekstrak daun jambu biji, kombinasi ekstrak dengan perbandingan 1:1, 2:1, dan 1:2. Hasil penelitian: ekstrak daun pepaya dan daun jambu biji baik tunggal maupun kombinasi mampu menghambat pertumbuhan bakteri Escherichia coli dengan diameter zona hambat ekstrak daun pepaya (11,21 mm), ekstrak daun jambu biji (10,69 mm), 1:1 (6,95 mm), 2:1 (7,10 mm) dan 1:2 (11,65 mm). Simpulan: Hasil diameter zona hambat tersebut dikategorikan sedang hingga kuat. Hasil uji efektivitas antibakteri oleh ekstrak daun pepaya sebesar 32,63%, ekstrak daun jambu biji sebesar 31,12%, 1:1 sebesar 20,23%, 2:1 sebesar 22,47%, dan 1:2 sebesar 33,91%. Dapat disimpulkan bahwa terdapat perbedaan efektivitas antibakteri antara ekstrak tunggal dan kombinasi daun pepaya dan daun jambu biji terhadap pertumbuhan bakteri Escherichia coli.
