Abstract
ANALYSIS OF LT AND ST GENES FROM Enterotoxigenic Escherichia coli BACTERIA USING POLYMERASE CHAIN REACTION METHOD ON LAWAR PLEK IN KETEWEL VILLAGE, GIANYAR ABSTRACT Lawar plek is a food made from raw meat. Raw meat potentially contain contaminant from pathogenic bacteria. One of the pathogenic bacteria is Enterotoxigenic Escherichia coli (ETEC). The purpose of this study is to identify Enterotoxigenic Escherichia coli bacteria using molecular method on lawar plek in Ketewel Village, Gianyar. This study use Eosin Methylene Blue Agar (EMBA) and continued by Polymerase Chain Reaction (PCR) as the identification method. The result form EMBA identification found 4 samples (33,33%) suspected colony of Escherichia coli bacteria and 8 samples (66,66%) neagitive. From that 4 samples, no virulence gene of LT (heat-labile toxin) and ST (heat-stable toxin) from Enterotoxigenic Escherichia coli are detected. From that results, the conclusion is that suspected Escherichia coli colony bacteria are found from the samples but none of them contain virulence gene of ETEC. Keywords: Enterotoxigenic Escherichia coli, lawar plek, polymerase chain reaction ANALISIS GEN LT DAN ST BAKTERI Enterotoxigenic Escherichia coli DENGAN METODE POLYMERASE CHAIN REACTION PADA LAWAR PLEK DI DESA KETEWEL, GIANYAR ABSTRAK Lawar plek adalah makanan yang dibuat dengan menggunakan daging mentah. Daging mentah berpotensi mengandung kontaminasi bakteri patogen. Salah satu bakteri tersebut adalah Enterotoxigenic Escherichia coli (ETEC). Tujuan dari penelitian ini adalah untuk mengidentifikasi secara molekuler bakteri Enterotoxigenic Escherichia coli pada makanan lawar plek di Desa Ketewel, Gianyar. Penelitian ini menggunakan metode identifikasi dengan Eosin Methylene Blue Agar (EMBA) dan dilanjutkan dengan identifikasi molekuler dengan Polymerase Chain Reaction (PCR). Hasil identifikasi EMBA ditemukan 4 sampel (33,33%) terduga koloni bakteri Escherichia coli. Dari 4 sampel tersebut, tidak ditemukan gen virulensi LT (heat-labile toxin) dan gen virulensi ST (heat-stable toxin) dari bakteri Enterotoxigenic Escherichia coli. Berdasarkan hasil tersebut dapat disimpulkan terdapat koloni terduga bakteri Eschericia coli pada sampel lawar plek, namun tidak ditemukan gen virulensi ETEC.
