Abstract

ABSTRACT Salmonella typhi is a pathogenic bacteria which causes typhoid fever. The infection that caused by Salmonella typhi can treat by giving antibiotics. At this time, it is reported that Salmonella typhi was resistant to antibiotics which have been effective for treatment. Therefore, it is necessary to develop natural-based antibiotics from various sources such as sembung leaves. This experiment’s purpose to observe the antibacterial activity on a various concentration of sembung leaves ethanol extracts toward Salmonella typhi bacteria growth by in vitro. Type of this research was true experiment designed with posttest only control design, using Kirby-Bauer diffusion disk method at four concentrations (20%, 40%, 60%, and 80%), group control (ethanol 96%) and work controls (Chloramphenicol antibiotic). Results showed that the sembung leaves ethanol extract was able to inhibits growth of Salmonella typhi with inhibition diameter zone of 20% (13,20 mm), 40% (14,22 mm), 60% (15,14 mm), and 80% (16,54 mm). These four concentration have strong inhibition potential. Based on the One Way Anova test showed the value of p < α (0,05) which means there were difference in growth inhibition zone of Salmonella typhi on various concentration of sembung leaves ethanol extracts and LSD test showed that there was a significant different on each concentration. In conclusion, this experiment provides that there is an antibacterial activity on a various concentration of sembung leaves ethanol extracts towards the growth of Salmonella typhi bacteria by in vitro. Keywords: sembung leaves ethanol extracts, Salmonella typhi, antibacterial activity, inhibition zone. ABSTRAK Salmonella typhi merupakan kuman patogen penyebab demam tifoid. Pengobatan terhadap infeksi oleh Salmonella typhi dilakukan dengan pemberian antibiotik. Saat ini dilaporkan Salmonella typhi mengalami resistensi terhadap antibiotik yang selama ini efektif untuk pengobatan. Oleh karena itu perlu dikembangkan antibiotik berbahan alam dari berbagai sumber seperti daun sembung. Penelitian ini bertujuan untuk mengetahui aktivitas antibakteri pada berbagai konsentrasi ekstrak etanol daun sembung terhadap pertumbuhan bakteri Salmonella typhi secara in vitro. Jenis penelitian yaitu true experiment dengan rancangan posttest only control design, menggunakan metode difusi cakram Kirby-Bauer pada empat konsentrasi 20%, 40%, 60% dan 80%, kelompok kontrol (etanol 96%) dan kontrol kerja (antibiotik Kloramfenikol). Hasil penelitian menunjukkan ekstrak etanol daun sembung mampu menghambat pertumbuhan Salmonella typhi dengan diameter zona hambat 20% (13,20 mm), 40% (14,22 mm), 60% (15,14 mm), dan 80% (16,54 mm). Keempat konsentrasi ini dikategorikan memiliki daya hambat yang kuat. Berdasarkan uji One Way Anova menunjukkan nilai p < α (0,05) sehingga ada perbedaan zona hambat pertumbuhan Salmonella typhi pada berbagai konsentrasi ekstrak etanol daun sembung dan uji LSD menunjukkan adanya perbedaan bermakna pada masing-masing konsentrasi. Simpulan penelitian ini adalah ada aktivitas antibakteri pada berbagai konsentrasi ekstrak etanol daun sembung terhadap pertumbuhan bakteri Salmonella typhi secara in vitro.
