Abstract
ANTIBACTERIAL ACTIVITY OF THE GAMAL LEAF (Gliricidia sepium) ETHANOL EXTRACT IN VARIOUS CONCENTRATION ON THE GROWTH of Streptococcus mutans BACTERIA IN VITRO ABSTRACT Background: Streptococcus mutans is positive gram bacteria which cause dental caries. Gamal leaf has active substance for antibacterial activity such as tannins, alkaloids, saponins and flavonoids. Purpose: The objective of the study was to know the antibacterial activity of ethanol extract of gamal leaf (Gliricidia sepium) for Streptococcus mutans in various concentrations. Method: This study is true experiment with post-test only control design. The determination of antibacterial activity in this study was conducted by using Kirby Bauer disk diffusion method with the various concentrations, there were 40%, 50%, 60%, 70% and 80%. The positive control in this study is 30 ìg disk diffusion chloramphenicols and the 96% of ethanol is used as the negative control. Result: The average inhibition of zone diameter in the concentrations of 40, 50, 60, 70 and 80% are 11.3 mm, 12.3 mm, 13.4 mm, 15.3 mm and 19.2 mm, respectively. One Way Anova statistic analysis showed that the value of p is 0.000, so there is a difference of growth inhibition zone of Streptococcus mutans in various concentrations of ethanol extract of gamal leaf (Gliriciadia sepium). Conclusion: Based on the result, it can be conclude that the ethanol extract of gamal leaf (Gliriciadia sepium) have strong antibacterial activity, especially to Streptococcus mutans bacteria. AKTIVITAS ANTIBAKTERI EKSTRAK ETANOL DAUN GAMAL (Gliricidia sepium) PADA BERBAGAI KONSENTRASI TERHADAP PERTUMBUHAN BAKTERI Streptococcus mutans SECARA IN-VITRO ABSTRAK Latar belakang Streptococcus mutans merupakan bakteri gram positif penyebab utama terjadinya karies gigi. Daun gamal mengandung zat aktif sekunder yang bersifat antibakteri seperti tanin, alkaloid, saponin, dan flavonoid. Tujuan penelitian ini bertujuan untuk mengetahui aktivitas antibakteri ekstrak Etanol daun gamal (Gliricidia sepium) pada berbagai konsentrasi terhadap pertumbuhan bakteri Streptococcus mutans secara in-vitro. Metode penelitian ini adalah true experimental dengan desain penelitian post-test only control design. Penentuan aktivitas antibakteri dilakukan dengan menggunakan metode difusi cakram Kirby Bauer dengan lima konsentrasi uji yaitu konsentrasi 40%, 50%, 60%, 70% dan 80%. Kontrol positif berupa cakram disk antibiotik Kloramfenikol 30 ìg dan kontrol negatif yaitu Etanol 96%. Hasil penelitian berupa rata-rata diameter zona hambat dari setiap konsentrasi yaitu konsentrasi 40%, 50%, 60%, 70% dan 80% dengan diameter zona hambat berturut-turut 11,3 mm, 12,3 mm, 13,4 mm, 15,3 mm dan 19,2 mm. Uji statistik One Way Anova menunjukkan nilai p adalah 0,000 sehingga ada perbedaan zona hambat pertumbuhan bakteri Streptococcus mutans pada berbagai konsentrasi ekstrak Etanol daun gamal (Gliricidia sepium). Simpulan dari penelitian ini ekstrak Etanol daun gamal (Gliriciadia sepium) memiliki aktivitas antibakteri yang kuat, terutama untuk bakteri Streptococcus mutans.
