Abstract

ANTIBACTERIAL ACTIVITY TEST OF COMBINATION OF NEEM AND LEGUNDI LEAF EXTRACTS AGAINST Staphylococcus aureus BACTERIA ABSTRACT Staphylococcus aureus is the most common bacteria causing infection with a prevalence of 36% which is resistant to antibiotics. Mimba leaves and legundi leaves contain flavonoids, alkaloids, saponins, tannins, steroids, triterpenoids which act as antibacterial substances. This study aims to determine the antibacterial activity of a combination extract of mimba and legundi leaves on the growth of S.aureus bacteria. This study was a pre-experimental design in the form of a "One-Shot Case Study" using the Kirby-Bauer disc diffusion method with four concentrations namely 20, 40, 60, and 80%, working controls using chloramphenicol 30 µg and negative controls using 96% ethanol. The results showed that the combined extracts of mimba and legundi leaves were able to inhibit S.aureus with an inhibition zone diameter of 20% by 3.08 mm±0,30, 40% by 4.33 mm±0,58, 60% by 5.95 mm±0,45, 80% by 6.95 mm±1,31. The LSD test showed a value of ρ˂α (0.05) so that there were differences in the growth inhibition zones of S.aureus between various concentrations of the combined extracts of mimba and legundi leaves. The conclusion of this study is that there are differences in the antibacterial activity of the combination extracts of mimba and legundi leaves on the growth of S.aureus bacteria. Daun mimba dan daun legundi mengandung senyawa flavonoid, alkaloid, saponin, tanin, steroid, triterpenoid yang berperan sebagai zat antibakteri. Penelitian ini bertujuan untuk mengetahui aktivitas antibakteri ekstrak kombinasi daun mimba dan daun legundi terhadap pertumbuhan bakteri S.aureus. Penelitian ini merupakan pre-experiment design dengan bentuk “One-Shot Case Study” menggunakan metode difusi cakram Kirby-Bauer dengan empat konsentrasi yaitu 20, 40, 60, dan 80%, kontrol kerja menggunakan kloramfenikol 30 µg dan kontrol negatif menggunakan etanol 96%. Hasil penelitian menunjukkan ekstrak kombinasi daun mimba dan daun legundi mampu menghambat S.aureus dengan diameter zona hambat 20% sebesar 3,08 mm±0,30, 40% sebesar 4,33 mm±0,58, 60% sebesar 5,95 mm±0,45, 80% sebesar 6,95 mm±1,31. Uji LSD menunjukkan nilai ρ˂α (0,05) sehingga ada perbedaan zona hambat pertumbuhan S.aureus antara berbagai konsentrasi ekstrak kombinasi daun mimba dan daun legundi. Simpulan penelitian ini adalah terdapat perbedaan aktivitas antibakteri ekstrak kombinasi daun mimba dan daun legundi terhadap pertumbuhan bakteri S.aureus.
