Abstract

IDENTIFICATION OF COLIFORM BACTERIA IN WELL WATER IN ANTIGA KELOD VILLAGE USING THE MOST PROBABLE NUMBER (MPN) METHOD ABSTRACT Background: Dug wells are still the main source of clean water for the community in Antiga Kelod Village, but they are vulnerable to contamination by coliform bacteria due to poor sanitation conditions. The presence of coliform bacteria can increase the risk of waterborne diseases such as diarrhea. Objective: This study aimed to determine the microbiological quality of dug well water through the identification of coliform bacteria using the Most Probable Number (MPN) method. Methods: This study used a descriptive design with 58 dug well water samples selected using stratified proportional random sampling. Examination was carried out using the 3-3-3 MPN method, and the results were compared with the Indonesian Ministry of Health Regulation No. 2 of 2023. Results: Most wells had a distance of less than 10 meters from septic tanks (59%), lacked a protective layer (55%), and had improperly sealed well covers (64%). The examination results showed that 32 samples (55%) did not meet the microbiological quality standards, with coliform values ranging from 0/100 mL to >1100/100 mL. Wells located close to septic tanks, without protective layers, and with improperly sealed covers tended to have water quality that did not meet the standards. Conclusion: Most dug well water in Antiga Kelod Village was contaminated with coliform bacteria and did not meet the microbiological standards for clean water quality. IDENTIFIKASI BAKTERI COLIFORM PADA AIR SUMUR GALI DI DESA ANTIGA KELOD DENGAN METODE MOST PROBABLE NUMBER (MPN) ABSTRAK Latar Belakang: Sumur gali masih menjadi sumber utama air bersih masyarakat di Desa Antiga Kelod, namun rentan tercemar bakteri coliform akibat kondisi sanitasi yang kurang baik. Keberadaan bakteri coliform dapat meningkatkan risiko penyakit berbasis air seperti diare. Tujuan: Penelitian ini bertujuan untuk mengetahui kualitas mikrobiologi air sumur gali melalui identifikasi bakteri coliform dengan metode Most Probable Number (MPN). Metode: Penelitian ini menggunakan desain deskriptif dengan jumlah sampel 58 air sumur gali yang dipilih menggunakan teknik stratified proportional random sampling. Pemeriksaan dilakukan menggunakan metode MPN ragam 3-3-3, kemudian hasil dibandingkan dengan Permenkes RI No. 2 Tahun 2023. Hasil: Sebagian besar sumur memiliki jarak dengan septiktank <10 meter (59%), dinding sumur tidak kedap air (59%), dan memiliki penutup tidak rapat (64%). Hasil pemeriksaan menunjukkan 32 sampel (55%) tidak memenuhi syarat kualitas mikrobiologi dengan nilai coliform berkisar 0/100 mL hingga >1100/100 mL. Sumur dengan jarak dekat terhadap septiktank, tanpa lapisan pelindung, dan penutup tidak rapat cenderung memiliki kualitas air yang tidak memenuhi syarat. Simpulan: Sebagian besar air sumur gali di Desa Antiga Kelod telah terkontaminasi bakteri coliform sehingga tidak memenuhi syarat kualitas mikrobiologi air bersih.
