Abstract
PHYTOCHEMICAL SCREENING AND ANTIBACTERIAL ACTIVITY OF GARLIC (Allium sativum) EXTRACT AGAINST Salmonella typhi ABSTRACT Background: Salmonella typhi is a pathogenic bacterium that causes typhoid fever and has shown increasing resistance to antibiotics. Garlic (Allium sativum) contains secondary metabolites with potential antibacterial properties. Objective: This study aimed to determine the phytochemical content and the inhibitory effect of garlic extract on the growth of Salmonella typhi. Methods: This study used an experimental method with a Posttest Only Control Group Design. The antibacterial activity was tested using the disc diffusion method at four extract concentrations: 15%, 35%, 55%, and 75%, with chloramphenicol as the positive control and 96% ethanol as the negative control. The data were analyzed using the One-Way ANOVA test and the LSD (Least Significant Difference) test. Results: Phytochemical screening showed that garlic extract contains flavonoids, alkaloids, and tannins, but tested negative for saponins. The inhibition test results indicated that no inhibition zones were formed at 15% and 35% concentrations, while inhibition zones were observed at 55% and 75% concentrations with average diameters of 7.75 mm and 9.37 mm, respectively. The positive control showed the largest inhibition zone (31.04 mm), while the negative control showed no inhibition. Statistical analysis using One Way ANOVA showed a significant value (p < 0.05), indicating differences between treatment groups. Conclusion: Garlic extract exhibits antibacterial activity against Salmonella typhi at certain concentrations, and its effectiveness increases with higher concentrations, supported by its phytochemical constituents. Keywords: Garlic, Phytochemical, Inhibition zone, Salmonella typhi UJI FITOKIMIA DAN AKTIVITAS ANTIBAKTERI EKSTRAK BAWANG PUTIH (Allium sativum) TERHADAP BAKTERI Salmonella typhi ABSTRAK Latar Belakang: Salmonella typhi merupakan bakteri patogen penyebab demam tifoid yang menunjukkan peningkatan resistensi terhadap antibiotik. Bawang putih (Allium sativum) mengandung senyawa metabolit sekunder yang berpotensi sebagai antibakteri. Tujuan: Penelitian ini bertujuan untuk mengetahui kandungan senyawa fitokimia serta daya hambat ekstrak bawang putih terhadap pertumbuhan bakteri Salmonella typhi. Metode: Penelitian ini menggunakan metode eksperimen dengan rancangan Posttest Only Control Group Design. Pengujian daya hambat dilakukan dengan metode difusi cakram pada empat konsentrasi ekstrak, yaitu 15%, 35%, 55%, dan 75%, dengan kloramfenikol sebagai kontrol positif dan etanol 96% sebagai kontrol negatif. Data dianalisis dengan uji One Way ANOVA dan LSD (Least Significant Difference). Hasil: Hasil uji fitokimia menunjukkan bahwa ekstrak bawang putih mengandung flavonoid, alkaloid, tanin, dan negatif saponin. Hasil uji daya hambat menunjukkan bahwa pada konsentrasi 15% dan 35% tidak terbentuk zona hambat, sedangkan pada konsentrasi 55% dan 75% terbentuk zona hambat dengan rata-rata diameter 7,75 mm dan 9,37 mm. Analisis statistik menggunakan uji One Way ANOVA menunjukkan nilai signifikan (p<0,05) yang berarti terdapat perbedaan antar kelompok perlakuan. Simpulan: Ekstrak bawang putih memiliki aktivitas antibakteri terhadap Salmonella typhi pada konsentrasi tertentu, dan efektivitasnya meningkat seiring peningkatan konsentrasi serta didukung oleh kandungan senyawa fitokimia yang dimilikinya.
