Abstract
INHIBITORY POWER TEST OF ETHANOL EXTRACT OF GEMITIR FLOWERS (Tagetes erecta Linn.) AGAINST Staphylococcus aureus USING DISC DIFFUSION METHOD ABSTRACT Background: Staphylococcus aureus infection remains a major health concern, increasingly complicated by the rise of methicillin-resistant S. aureus (MRSA) and antibiotic resistance. Misuse of antibiotics accelerates resistance, highlighting the need for alternative antibacterial agents from natural sources. Marigold (Tagetes erecta L.) contains bioactive compounds such as flavonoids, phenolics, and alkaloids with potential antibacterial activity. Purpose: This study aimed to evaluate the inhibitory effect of 70% ethanol extract of marigold flower on the growth of Staphylococcus aureus. Method: A true experimental design with a posttest-only control group was used. The antibacterial activity was assessed using the disc diffusion method at extract concentrations of 5%, 10%, 15%, and 20%, with four replications. Data were analyzed using Shapiro–Wilk, Levene’s test, One Way ANOVA (Welch), and Games–Howell post hoc test. Results: The mean inhibition zone diameters at concentrations of 5%, 10%, 15%, and 20% were 21.65 mm, 24.12 mm, 24.58 mm, and 28.92 mm, respectively, all categorized as very strong. Statistical analysis showed significant differences among concentrations (p ≤ 0.05). The 20% concentration showed the highest inhibitory effect. Conclusion: The 70% ethanol extract of marigold flower has strong potential as a natural antibacterial agent against Staphylococcus aureus. Keywords: Marigold, Inhibition zone, Staphylococcus aureus. UJI DAYA HAMBAT EKSTRAK ETANOL BUNGA GEMITIR (Tagetes erecta Linn.) TERHADAP Staphylococcus aureus DENGAN METODE DIFUSI CAKRAM ABSTRAK Latar Belakang: Infeksi Staphylococcus aureus masih menjadi masalah kesehatan utama yang semakin kompleks akibat meningkatnya prevalensi methicillin-resistant Staphylococcus aureus serta resistensi antibiotik. Penggunaan antibiotik yang tidak tepat mempercepat resistensi sehingga diperlukan alternatif antibakteri dari bahan alam. Salah satu tanaman yang berpotensi adalah bunga gemitir (Tagetes erecta L.) yang mengandung metabolit sekunder seperti flavonoid, fenolik, dan alkaloid. Tujuan: penelitian ini bertujuan untuk mengetahui daya hambat ekstrak etanol 70% bunga gemitir terhadap pertumbuhan Staphylococcus aureus. Metode: penelitian ini menggunakan true experimental dengan desain posttest-only control group. Uji daya hambat dilakukan dengan metode difusi cakram pada konsentrasi ekstrak 5%, 10%, 15%, dan 20% dengan dilakukan pengulangan sebanyak empat kali. Data dianalisis menggunakan uji Shapiro-Wilk, uji Levene, uji One Way ANOVA (Welch), dan uji post hoc Games-Howell. Hasil: penelitian menunjukkan bahwa rata-rata diameter zona hambat pada konsentrasi 5%, 10%, 15%, dan 20% berturut-turut sebesar 21,65 mm; 24,12 mm; 24,58 mm; dan 28,92 mm, yang seluruhnya termasuk kategori sangat kuat. Analisis statistik menunjukkan adanya perbedaan signifikan antar konsentrasi (p ≤ 0,05). Konsentrasi 20% menghasilkan daya hambat terbesar terhadap pertumbuhan Staphylococcus aureus. Simpulan: Ekstrak etanol 70% bunga gemitir memiliki daya hambat yang sangat kuat terhadap Staphylococcus aureus dan berpotensi sebagai alternatif agen antibakteri alami.
