Abstract

ANTIBACTERIAL ACTIVITY TEST OF GOTU KOLA (CENTELLA ASIATICA L.) LEAF EXTRACT BASED ON DIFFERENT SOLVENTS AGAINST STAPHYLOCOCCUS AUREUS BACTERIA ABSTRACT Background: Bacterial infections remain a significant health problem, one of which is caused by Staphylococcus aureus. Inappropriate use of antibiotics may lead to resistance; therefore, alternative antibacterial agents from natural sources such as gotu kola (Centella asiatica L.) are needed. Solvent type may influence the extraction of active compounds and antibacterial activity. Purpose: This study aimed to determine differences in antibacterial activity of gotu kola leaf extract using ethanol 96%, ethyl acetate, and n-hexane against Staphylococcus aureus. Methods: This true experimental study used a posttest only control group design. Antibacterial activity was tested in vitro using the disc diffusion method on MHA with 50% extract concentration and five repetitions. Data were analyzed using SPSS with Shapiro-Wilk, Levene, One Way ANOVA, and Tukey HSD tests. Results: Mean inhibition zones were 10.75 mm (ethanol, strong), 7.70 mm (ethyl acetate, moderate), and 5.72 mm (n-hexane, moderate) based on Davis & Stout classification. Data were normally distributed and homogeneous (p>0.05). ANOVA showed significant differences (p=0.001). Tukey test indicated ethanol extract had significantly higher activity than the others (p<0.05). Conclusion: Antibacterial activity differs by solvent type, with ethanol extract showing the highest effectiveness. BERDASARKAN PERBEDAAN PELARUT TERHADAP BAKTERI Staphylococcus aureus ABSTRAK Latar Belakang: Infeksi bakteri masih menjadi masalah kesehatan yang signifikan, salah satunya disebabkan oleh Staphylococcus aureus. Penggunaan antibiotik yang tidak tepat dapat memicu resistensi, sehingga diperlukan alternatif antibakteri dari bahan alam seperti daun pegagan (Centella asiatica L.). Perbedaan jenis pelarut dapat memengaruhi kandungan senyawa aktif dan aktivitas antibakteri. Tujuan: Penelitian ini bertujuan untuk mengetahui perbedaan aktivitas antibakteri ekstrak daun pegagan dengan pelarut etanol 96%, etil asetat, dan n-heksana terhadap Staphylococcus aureus. Metode: Penelitian ini merupakan true experimental dengan rancangan posttest only control group design. Uji dilakukan secara in vitro menggunakan metode difusi cakram pada media MHA dengan konsentrasi 50% dan lima kali pengulangan. Analisis data menggunakan uji Shapiro-Wilk, Levene, One Way ANOVA, dan Tukey HSD. Hasil: Rerata zona hambat etanol 10,75 mm (kuat), etil asetat 7,70 mm (sedang), dan n-heksana 5,72 mm (sedang) berdasarkan Davis & Stout. Data berdistribusi normal dan homogen (p>0,05). ANOVA menunjukkan perbedaan signifikan (p=0,001). Uji Tukey menunjukkan etanol lebih tinggi signifikan dibanding lainnya (p<0,05). Simpulan: Aktivitas antibakteri berbeda berdasarkan pelarut, dengan etanol sebagai yang paling efektif. Kata kunci: Centella asiatica L., antibakteri
