Abstract
ANTIBACTERIAL ACTIVITY TEST OF MENIRAN LEAF EXTRACT (Phyllanthus sp.) ON THE GROWTH OF Staphylococcus aureus BACTERIA ABSTRACT Background Infections caused by Staphylococcus aureus infection remain a major health problem due to increasing antibiotic resistance, therefore alternative antibacterial agents from natural sources such as meniran leaves (Phyllanthus sp.) are needed. Objective This study aimed to determine the antibacterial activity of meniran leaf extract against the growth of Staphylococcus aureus. Methods This laboratory experimental study used the disk diffusion method with extract concentrations of 20%, 40%, 60%, and 80%, each tested in six replications. The observed parameter was the inhibition zone diameter, followed by phytochemical screening. Data were analyzed using the One Way ANOVA test. Results showed that meniran leaf extract contained alkaloids, flavonoids, tannins, saponins, and terpenoids. All extract concentrations inhibited the growth of Staphylococcus aureus with mean inhibition zone diameters of 7.57 ± 0.21 mm, 8.80 ± 0.24 mm, 10.34 ± 0.31 mm, and 11.70 ± 0.34 mm, respectively. The 80% concentration produced the largest inhibition zone, and statistical analysis showed a significant difference among concentrations (p<0.05). Conclusion meniran leaf extract has antibacterial activity against Staphylococcus aureus, and higher extract concentrations produce greater inhibitory effects. TERHADAP PERTUMBUHAN BAKTERI Staphylococcus aureus ABSTRAK Latar Belakang Infeksi yang disebabkan oleh bakteri Staphylococcus aureus masih menjadi masalah kesehatan akibat meningkatnya resistensi antibiotik, sehingga diperlukan alternatif antibakteri dari bahan alam seperti daun meniran (Phyllanthus sp.). Tujuan penelitian ini bertujuan untuk mengetahui aktivitas antibakteri ekstrak daun meniran terhadap pertumbuhan bakteri Staphylococcus aureus. Metode penelitian ini merupakan penelitian eksperimental laboratorium dengan metode difusi cakram menggunakan variasi konsentrasi ekstrak 20%, 40%, 60%, dan 80% dengan enam kali pengulangan. Parameter yang diamati adalah diameter zona hambat serta dilakukan skrining fitokimia. Data dianalisis menggunakan uji One Way ANOVA. Hasil penelitian menunjukkan bahwa ekstrak daun meniran mengandung alkaloid, flavonoid, tanin, saponin, dan terpenoid. Seluruh konsentrasi ekstrak mampu menghambat pertumbuhan Staphylococcus aureus dengan rata-rata diameter zona hambat berturut-turut sebesar 7,57 ± 0,21 mm; 8,80 ± 0,24 mm; 10,34 ± 0,31 mm; dan 11,70 ± 0,34 mm. Konsentrasi 80% menghasilkan zona hambat terbesar dan hasil uji statistik menunjukkan terdapat perbedaan signifikan antar konsentrasi (p<0,05). Simpulan penelitian ini adalah ekstrak daun meniran memiliki aktivitas antibakteri terhadap Staphylococcus aureus dan semakin tinggi konsentrasi ekstrak maka semakin besar daya hambat yang dihasilkan..
