Abstract

POLYMORPHISM OF THE Melatonin Receptor 1B GENE AS A RISK FACTOR FOR TYPE 2 DIABETES MELLITUS IN OBESITY CASES USING THE PCR-RFLP METHOD ABSTRACT Background: Type 2 diabetes mellitus is a chronic metabolic disease with increasing global prevalence, particularly among individuals with obesity. Inaddition to environmental factors such as diet, physical activity, and sleep patterns, genetic factors also play a crucial role, one of which is the MTNR1B gene that regulates circadian rhythm and insulin secretion. Objective: to determine the polymorphism of the MTNR1B gene as a risk factor for type 2 diabetes mellitus in obesity cases using the PCR-RFLP method. Methods: This descriptive study involved 10 obese and 10 non-obese respondents selected through purposive sampling (accidental). Blood samples were collected for DNA extraction, followed by amplification of the target gene using specific primers and digestion using restriction enzymes. Results: Electrophoresis results revealed a single DNA band at 980 bp in all samples, indicating that all respondents had a homozygous genotype with no allele variations. Conclusion: Based on these findings, it can be concluded that no MTNR1B gene polymorphism was found in the study population. Further research with a larger sample size and advanced molecular techniques is needed to better understand the role of this gene in the risk of developing type 2 diabetes mellitus among obese individuals. Keywords: Type 2 diabetes mellitus, Obesity, MTNR1B gene, PCR-RFLP POLIMORFISME GEN Melatonin Receptor 1B SEBAGAI FAKTOR RISIKO KEJADIAN DIABETES MELITUS TIPE II PADA KASUS OBESITAS DENGAN MENGGUNAKAN METODE PCR-RFLP ABSTRAK Latar Belakang: Diabetes melitus tipe 2 merupakan penyakit metabolik kronis yang terus meningkat prevalensinya, terutama pada individu dengan obesitas. Selain faktor lingkungan seperti pola makan, aktivitas fisik, dan pola tidur, faktor genetik juga memegang peran penting, salah satunya adalah gen MTNR1B yang berperan dalam regulasi ritme sirkadian dan sekresi insulin. Tujuan: Penelitian ini bertujuan untuk mengetahui polimorfisme gen MTNR1B sebagai faktor risiko kejadian diabetes melitus tipe 2 pada kasus obesitas menggunakan metode PCRRFLP. Metode: Penelitian ini merupakan penelitian deskriptif dengan teknik purposive sampling jenis sampling aksidental erhadap 10 responden obesitas dan 10 responden non-obesitas. Sampel darah diambil dan dilakukan ekstraksi DNA, dilanjutkan dengan amplifikasi gen menggunakan primer spesifik serta digesti oleh enzim restriksi. Hasil: Hasil elektroforesis menunjukkan semua sampel memiliki satu pita DNA berukuran 980 bp, yang menunjukkan bahwa seluruh responden memiliki genotipe homozigot tanpa adanya variasi alel. Simpulan: Berdasarkan hasil tersebut, dapat disimpulkan bahwa pada populasi sampel ini tidak ditemukan polimorfisme gen MTNR1B. Diperlukan penelitian lanjutan dengan sampel yang lebih luas dan teknik analisis molekuler lanjutan untuk mengeksplorasi lebih dalam hubungan gen ini terhadap risiko diabetes melitus tipe 2 pada individu obesitas.
