Abstract

Background: Staphylococcus aureus is one of the most common bacterial infections in the world. In addition, there has been an increase in the resistance of these bacteria to various types of antibiotics (multi drug resistance). Research has proven that betel (Piper betle L.) has the potential to be antibacterial because it has a variety of active compounds. Betel leaf can be processed into extracts and decoction. The purpose of this study was to determine the inhibitory activity of ethanol extract and decoction of betel leaf (Piper betle L.) on the growth of Staphylococcus aureus by using the agar dilution method. Methods: This type of research is a True Experimental Research, with the research design of the Posttest Equivalent Groups Study. Antibacterial test for bacteria used the agar dilution test method with various concentrations of 20, 25, 35, 50%. The positive control used was amoxicillin antibiotic and the negative control used aquades and ethanol. Results: The decoction of betel leaves decreased the colony of Staphylococcus aureus bacteria significantly at a concentration of 20, 25, and 35%, while at a concentration of 50% there were no more Staphylococcus aureus colonies growing. The ethanol extract of betel leaf had no Staphylococcus aureus bacteria colonies that grew at concentrations of 20, 25, 35, and 50%. Continued with the Kruskal Wallis test which showed a significant difference from the number of Staphylococcus aureus colonies that grew from the decoction of betel leaves at concentrations of 20, 25, 35, and 50%. Whereas in the ethanol extract of the betel leaf there was no significant difference from the number of bacterial colonies that grew from the ethanol extract of betel leaf at concentrations of 20, 25, 35, and 50%. Conclusion: The ethanol extract and decoction of betel leaf can inhibit the growth of Staphylococcus aureus, where the Minimum Bactericidal Concentration (MBC) and Minimum Inhibitory Concentration (MIC) of the betel leaf ethanol extract cannot be determined because at the lowest concentration (20%) there is no colony growth. Staphylococcus aureus, while the decoction of betel leaves shows a Minimum Inhibitory Concentration (MIC) at a concentration of 20%, and a Minimum Bactericidal Concentration (MBC) at a concentration of 50%. So it is necessary to carry out further research on the ethanol extract of betel leaves with a concentration lower than 20% to determine the Minimum Bactericidal Concentration (MBC) and Minimum Inhibitory Concentration (MIC).
