Abstract

ABSTRACT Background : Acne is a skin disease that occurs due to a blockage of sebaceous follicles caused by Propionibacterium acnes. Legundi leaf contain alkaloids, flavonoids, tannins, saponins and steroids as antibacteria. Objective : This study was conducted to determine the differences in inhibition of the combination of legundi leaf extract (Vitex trifolia L.) and virgin coconut oil with various concentrations of 15%, 20%, 30%, 45% and 60% against Propionibacterium acnes bacteria. Method : This study uses a type of research with a One-Shot Case Study design with disc diffusion method. This study uses five concentrations, namely 15%, 20%, 30%, 45% and 60%, and research controls using virgin coconut oil and amoxicillin antibiotics as work controls. The inhibition zone is determined by looking at the clear zone found around the disc. The statistical analysis used was the Kolmogorov-Smirnov test, One Way Anova, and the Least Significant Deference test. Result: The results showed the average diameter of the inhibition zone produced a combination of legundi leaf and virgin coconut oil with concentrations of 15%, 20%, 30%, 45% and 60%, namely 7.2 mm, 7.5 mm, 7.45 mm, 8 .5 mm and 10.25 mm. Based on the One Way Anova and Least Significant Deference tests, it is known that there are differences in the diameter of inhibition zones in the growth of Propionibacterium acnes in various concentrations of the combination of legundi leaf and virgin coconut oil. Conclusion: The combination of legundi leaf and virgin coconut oil can inhibit the growth of Propionibacterium acnes bacteria and there are significant differences in the diameter of the inhibitory zone of Propionibacterium acnes growth in various concentrations. Keyword : Legundi leaves, virgin coconut oil, Propionibacterium acnes, inhibitory power ABSTRAK Latar belakang: Jerawat merupakan penyakit kulit yang terjadi karena adanya penyumbatan folikel sebasea yang disebabkan oleh bakteri Propionibacterium acnes. Daun legundi mengandung alkaloid, flavonoid, tanin, saponin dan steroid sebagai antibakteri. Tujuan: Penelitian ini dilakukan untuk mengetahui perbedaan daya hambat kombinasi ekstrak daun legundi (Vitex trifolia L.) dan virgin coconut oil dengan berbagai konsentrasi 15%, 20%, 30%, 45% dan 60% terhadap bakteri Propionibacterium acnes. Metode: Penelitian ini menggunakan jenis penelitian dengan rancangan penelitian One-Shot Case Study dengan metode difusi cakram. Penelitian ini menggunakan lima konsentrasi yaitu 15%, 20%, 30%, 45% dan 60%, dan terdapat kontrol penelitian yang menggunakan virgin coconut oil serta antibiotik amoxicillin sebagai kontrol kerja. Zona hambat ditentukan dengan melihat zona bening yang terdapat di sekeliling cakram. Analisis statistik yang digunakan adalah uji Kolmogorov-Smirnov, One Way Anova, dan uji Least Significant Deference. Hasil: Hasil menunjukkan rata-rata diameter zona hambat yang dihasilkan kombinasi daun legundi dan virgin coconut oil dengan konsentrasi 15%, 20%, 30%, 45% dan 60% yaitu 7,2 mm, 7,5 mm, 7,45 mm, 8,05 mm dan 10,25 mm. Berdasarkan uji One Way Anova dan Least Significant Deference diketahui terdapat perbedaan diameter zona hambat pada pertumbuhan bakteri Propionibacterium acnes pada berbagai variasi konsentrasi kombinasi daun legundi dan virgin coconut oil. Simpulan: Kombinasi daun legundi dan virgin coconut oil mampu menghambat pertumbuhan bakteri Propionibacterium acnes dan terdapat perbedaan diameter zona hambat pertumbuhan bakteri Propionibacterium acnes yang signifikan pada berbagai variasi konsentrasi.
