Abstract

ANALYSIS QUALITATIVE OF HEPATITIS B VIRUS DNA FROM HEPATITIS B SURFACE ANTIGEN POSITIF WITH POLYMERASE CHAIN REACTION TECHNIQUE ABSTRACT The infection by the hepatitis B virus is starting to be a big and serious public health problem. Hepatitis B is caused by the hepatitis B virus, a part of the hepadnavirus family that can cause acute or chronic inflammation of the liver and can become to liver cirrhosis or liver cancer. The purposed of this research is to analyze hepatitis B virus DNA from positive HBsAg blood samples by PCR technique. The type of this research is descriptive. Plasma samples were taken at the Wangaya Regional General Hospital, totaling 10 samples. Plasma samples were isolated using Qiagen reagent kits. Then processed with the PCR test to see the DNA fragments produced. The results of this research shows that the results of the PCR test using 251f primers and 1190r primers indicate that the presence of DNA bands that fluoresce between the 4th and 5th markers with molecular weights between 800-1000bp. The presence of fluorescent DNA bands shows that the primer is specific to the hepatitis B virus which mean the results are directly proportional to the primary set used. Hepatitis B disebabkan oleh virus hepatitis B, suatu anggota famili hepadnavirus yang dapat menyebabkan peradangan hati akut atau menahun dan dapat berlanjut menjadi sirosis hati atau kanker hati. Tujuan dari penelitian ini adalah untuk menganalisis DNA virus hepatitis B dari sampel darah HBsAg positif dengan teknik PCR. Penelitian ini merupakan penelitian deskriptif. Sampel plasma diambil di Rumah Sakit Umum Daerah Wangaya yang berjumlah 10 sampel. Sampel plasma diisolasi menggunakan kit reagen Qiagen. Kemudian dilanjutkan dengan uji PCR untuk melihat fragmen DNA yang dihasilkan. Hasil penelitian ini menunjukkan bahwa hasil uji PCR dengan menggunakan primer 251f dan primer 1190r menunjukkan adanya hasil pita DNA yang berpendar diantara marker ke-4 dan ke-5 dengan berat molekul antara 800-1000bp. Adanya pita DNA yang berpendar menunjukkan bahwa primer tersebut spesifik terhadap virus hepatitis B dimana hasil tersebut berbanding lurus dengan set primer yang digunakan. Adanya pita DNA yang berpendar menunjukkan bahwa primer yang digunakan spesifik terhadap virus hepatitis B. Kata kunci: PCR, Virus Hepatitis B, DNA, HBsAg
