Abstract

ANTIBACTERIAL ACTIVITY TEST OF Staphylococcus aureus BASED ON DIFFERENT TYPES OF SOLVENTS FROM BILIMBI LEAF EXTRACT (Averrhoa bilimbi L.) ABSTRACT Bilimbi leaves (Averrhoa bilimbi L.) are known to contain various secondary metabolites with potential antibacterial properties. This study aimed to determine the secondary metabolite compounds and antibacterial activity of bilimbi leaf extract against Staphylococcus aureus based on different extraction solvents. The extraction method used was maceration with three solvents: water, 70% ethanol, and n-hexane. Phytochemical screening was conducted to identify secondary metabolites, while antibacterial activity was evaluated using the disc diffusion method with four replications. Data were analyzed using Kruskal–Wallis and Mann–Whitney tests. The phytochemical screening results showed that the water extract contained tannins, the 70% ethanol extract contained flavonoids and tannins, while the n-hexane extract contained flavonoids and terpenoids. Alkaloids and saponins were not detected in all extracts. The antibacterial test showed that the 70% ethanol extract inhibited the growth of Staphylococcus aureus with an average inhibition zone diameter of 8.77 mm, whereas the water and n-hexane extracts showed no inhibition zone (0 mm). Statistical analysis indicated a significant difference among treatments (p<0.05). It can be concluded that the 70% ethanol extract of bilimbi leaves possesses antibacterial activity against Staphylococcus aureus. UJI AKTIVITAS ANTIBAKTERI Staphylococcus aureus BERDASARKAN PERBEDAAN JENIS PELARUT EKSTRAK DAUN BELIMBING WULUH (Averrhoa bilimbi L.) ABSTRAK Daun belimbing wuluh (Averrhoa bilimbi L.) diketahui mengandung berbagai senyawa metabolit sekunder yang berpotensi sebagai antibakteri. Penelitian ini bertujuan untuk mengetahui kandungan metabolit sekunder serta aktivitas antibakteri ekstrak daun belimbing wuluh terhadap bakteri Staphylococcus aureus berdasarkan perbedaan jenis pelarut ekstraksi. Metode ekstraksi yang digunakan adalah maserasi dengan tiga jenis pelarut, yaitu air, etanol 70%, dan n-heksana. Skrining fitokimia dilakukan untuk mengidentifikasi kandungan metabolit sekunder, sedangkan uji aktivitas antibakteri dilakukan menggunakan metode difusi cakram dengan empat kali pengulangan. Data dianalisis menggunakan uji Kruskal–Wallis dan Mann–Whitney. Hasil skrining fitokimia menunjukkan bahwa ekstrak air mengandung tanin, ekstrak etanol 70% mengandung flavonoid dan tanin, sedangkan ekstrak n-heksana mengandung flavonoid dan terpenoid. Senyawa alkaloid dan saponin tidak terdeteksi pada seluruh ekstrak. Hasil uji antibakteri menunjukkan bahwa ekstrak etanol 70% mampu menghambat pertumbuhan Staphylococcus aureus dengan rata-rata diameter zona hambat sebesar 8,77 mm, sedangkan ekstrak air dan n-heksana tidak menunjukkan zona hambat (0 mm). Uji statistik menunjukkan terdapat perbedaan signifikan antar perlakuan (p<0,05). Disimpulkan bahwa ekstrak etanol 70% daun belimbing wuluh memiliki aktivitas antibakteri terhadap Staphylococcus aureus.
