Abstract
ANTIFUNGAL ACTIVITY TEST OF GREEN EXTRACTION VCO OF ENCOK ROOTS (Plumbago zeylanica L.) AGAINST Candida albicans FUNGI ABSTRACT Background: Candida albicans is an opportunistic pathogenic fungus that causes candidiasis with a prevalence of around 20-25% of cases in Indonesia and is starting to show resistance to several synthetic antifungal drugs. This encourages the search for safer and more effective alternative therapies. Encok roots (Plumbago zeylanica L.) are known to contain secondary metabolites that have the potential to act as antifungals, and the green extraction method using Virgin Coconut Oil (VCO) as a solvent was chosen because it is environmentally friendly. Purpose: This study aims to determine the antifungal activity of encok root extracts from green extraction using VCO against the growth of Candida albicans by determining the Minimum Inhibitory Concentration (MIC). Methods: This study is a true experimental study using the liquid dilution method using six concentrations (15%, 7.5%, 3.75%, 1.88%, 0.94%, 0.47%), as well as positive, negative, and solvent controls. Inhibition of fungal growth was determined by measuring the absorbance before and after incubation using spectrophotometry. Results: A concentration of 15% showed the highest inhibitory power, while 0.47% did not provide any inhibitory effect. A concentration of 1.88% was the lowest concentration that showed consistent inhibitory power and was significantly different (p<0.05) from the negative control. Conclusion: The MIC of Plumbago zeylanica root extract from green extraction using VCO against Candida albicans was at a concentration of 1.88%. Keywords: gout root; MIC; Candida albicans. TERHADAP JAMUR Candida albicans ABSTRAK Latar belakang: Candida albicans merupakan jamur patogen oportunistik penyebab kandidiasis dengan pravelensi sekitar 20-25% kasus di Indonesia serta mulai menunjukkan resistensi terhadap beberapa obat antijamur sintesis. Hal ini mendorong pencarian alternatif terapi yang lebih aman dan efektif. Akar encok (Plumbago zeylanica L.) diketahui mengandung metabolit sekunder berpotensi sebagai antijamur, dan metode green extraction dengan pelarut Virgin Coconut Oil (VCO) dipilih karena ramah lingkungan. Tujuan: Penelitian ini bertujuan untuk mengetahui aktivitas antijamur ekstrak akar encok hasil green extraction menggunakan VCO terhadap pertumbuhan Candida albicans melalui penentuan Konsentrasi Hambat Minimum (KHM). Metode: Penelitian ini merupakan true experimental dengan metode dilusi cair menggunakan enam konsentrasi (15%, 7,5%, 3,75%, 1,88%, 0,94%, 0,47%), serta kontrol positif, negatif dan pelarut. Penghambatan pertumbuhan jamur ditentukan melalui pengukuran absorbansi sebelum dan sesudah inkubasi secara speaktrofotometri. Hasil: Konsentrasi 15% menunjukkan daya hambat tertinggi, sedangkan 0,47% tidak memberikan efek penghambatan. Konsentrasi 1,88% merupakan konsentrasi terendah yang menunjukkan daya hambat konsisten dan berbeda signifikan (p<0,05) dengan kontroll negatif. Simpulan: KHM ekstrak akar encok (Plumbago zeylanica L.) hasil green extraction menggunakan VCO terhadap Candida albicans yaitu pada konsentrasi 1,88%. Kata kunci : akar encok; KHM; Candida albicans.
