Abstract
ANTIBACTERIAL ACTIVITY TEST OF COCONUT PALM WINE AGAINST THE GROWTH OF Staphylococcus aureus ABSTRAK Background: Staphylococcus aureus is a pathogenic bacterium that can cause various infections and has shown increasing resistance to antibiotics. Therefore, alternative antibacterial agents from natural sources are needed. Fermented coconut palm wine is known to contain bioactive compounds such as ethanol, organic acids, and phytochemical compounds that have potential antibacterial properties. Objective: this study aimed to determine the antibacterial activity of coconut palm wine against the growth of Staphylococcus aureus. Methods: his study was an experimental study using a Posttest Only Control Group Design and the disc diffusion method. The sample used was coconut palm wine fermented for 7 days with concentration variations of 20%, 30%, 40%, and 50%. The positive control used gentamicin, while the negative control used distilled water (aquades). Results: The results showed that coconut palm wine was able to inhibit the growth of Staphylococcus aureus, with inhibition zone diameters of 7.05 mm at 20% concentration, 8.59 mm at 30%, 9.43 mm at 40%, and 10.45 mm at 50%. Statistical analysis using One Way ANOVA showed a p-value of <0.05, indicating a significant difference among the concentrations. Conclusion: coconut palm wine has antibacterial activity against the growth of Staphylococcus aureus, and its effectiveness increases with higher concentrations, indicating its potential as a natural antibacterial alternative. UJI AKTIVITAS ANTIBAKTERI MINUMAN TUAK KELAPA TERHADAP PERTUMBUHAN BAKTERI Staphylococcus aureus ABSTRAK Latar Belakang: Staphylococcus aureus merupakan bakteri patogen yang dapat menyebabkan berbagai infeksi dan menunjukkan peningkatan resistensi terhadap antibiotik. Oleh karena itu, diperlukan alternatif antibakteri dari bahan alami. Minuman tuak kelapa hasil fermentasi diketahui mengandung senyawa bioaktif seperti etanol, asam organik, dan senyawa fitokimia yang berpotensi sebagai antibakteri. Tujuan: penelitian ini bertujuan untuk mengetahui aktivitas antibakteri minuman tuak kelapa terhadap pertumbuhan Staphylococcus aureus. Metode: penelitian ini merupakan penelitian eksperimental dengan rancangan Posttest Only Control Group Design menggunakan metode difusi cakram. Sampel yang digunakan adalah tuak kelapa hasil fermentasi selama 7 hari dengan variasi konsentrasi 20%, 30%, 40%, dan 50%, serta kontrol positif menggunakan antibiotic gentamicin dan kontrol negatif menggunakan aquades. Hasil: penelitian menunjukkan bahwa tuak kelapa mampu menghambat pertumbuhan Staphylococcus aureus, dengan diameter zona hambat pada konsentrasi 20% sebesar 7,05 mm, konsentrasi 30% sebesar 8,59 mm, konsentrasi 40% sebesar 9,43 mm dan konsentrasi 50% sebesar 10,45%. Uji statistik One Way ANOVA menunjukkan nilai p<0,05 yang berarti terdapat perbedaan bermakna antar konsentrasi. Simpulan: minuman tuak kelapa memiliki aktivitas antibakteri terhadap pertumbuhan Staphylococcus aureus dan efektivitasnya meningkat seiring dengan peningkatan konsentrasi, sehingga berpotensi sebagai alternatif antibakteri alami.
