Abstract
ANTIBACTERIAL ACTIVITY TEST OF CHINESE BETEL LEAF AND STEM EXTRACT (Peperomia sp.) ON THE GROWTH OF Staphylococcus aureus BASED ON THE DETERMINATION OF THE MINIMUM INHIBITORY CONCENTRATION ABSTRACT Background: Skin infections, particularly those caused by Staphylococcus aureus, remain a significant health problem exacerbated by increasing antibiotic resistance, thereby necessitating alternative antibacterial agents derived from natural sources. Objective: To determine the antibacterial activity of leaf and stem extracts of sirih cina (Peperomia sp.), identify the Minimum Inhibitory Concentration (MIC), and analyze differences in effectiveness across various concentrations. Methods: Laboratory experimental study using a pretest-posttest control group design. The extract was obtained through maceration using 96% ethanol and tested using the broth dilution method against Staphylococcus aureus ATCC 29213 at concentrations of 50%, 25%, 12.5%, 6.25%, 3.125%, 1.56%, 0.78%, and 0.39%. Antibacterial activity was assessed based on changes in optical density (OD). Results: Showed a decrease in OD at concentrations of 50%, 25%, 12.5%, 6.25%, and an increase in OD at concentrations of 3.125%, 1.56%, 0.78%, 0.39%. The MIC was determined at 6.25%, representing the lowest concentration that effectively inhibited bacterial growth. Welch ANOVA showed p = 0.000 (p < 0.05), and the Games-Howell Post Hoc test indicated significant differences between certain concentration groups (p < 0.05). Conclusion: Peperomia sp. leaf and stem extract exhibits antibacterial activity against Staphylococcus aureus, with effectiveness increasing proportionally with concentration. TERHADAP PERTUMBUHAN BAKTERI Staphylococcus aureus BERDASARKAN PENENTUAN KONSENTRASI HAMBAT MINIMUM ABSTRAK Latar Belakang: Infeksi kulit, khususnya oleh Staphylococcus aureus, masih menjadi masalah kesehatan yang diperburuk oleh peningkatan resistensi antibiotik sehingga diperlukan alternatif antibakteri berbahan alam. Tujuan: Mengetahui aktivitas antibakteri ekstrak daun dan batang sirih cina (Peperomia sp.), menentukan Konsentrasi Hambat Minimum (KHM), serta menganalisis perbedaan efektivitas pada berbagai konsentrasi. Metode: Eksperimen laboratorium dengan desain pretest-posttest control group. Ekstrak diperoleh melalui metode maserasi menggunakan etanol 96% dan diuji dengan metode dilusi cair terhadap Staphylococcus aureus ATCC 29213 pada konsentrasi 50%, 25%, 12,5%, 6,25%, 3,125%, 1,56%, 0,78%, dan 0,39%. Aktivitas antibakteri diukur berdasarkan perubahan nilai optical density (OD). Hasil: Menunjukkan penurunan OD pada konsentrasi 50%, 25%, 12,5%, 6,25% dan peningkatan OD pada konsentrasi 3,125%, 1,56%, 0,78%, 0,39%. KHM ditetapkan pada konsentrasi 6,25% yaitu konsentrasi terendah yang masih mampu menghambat pertumbuhan bakteri secara nyata. Uji Welch ANOVA menunjukkan p = 0,000 (p < 0,05) dan uji Games-Howell Post Hoc menunjukkan perbedaan signifikan antar konsentrasi tertentu (p < 0,05). Simpulan: Ekstrak daun dan batang sirih cina (Peperomia sp.) memiliki aktivitas antibakteri terhadap Staphylococcus aureus yang meningkat seiring dengan konsentrasi.
