Abstract

PERBEDAAN NILAI ACTIVATED PARTIAL THROMBOPLASTIN TIME (APTT) PLASMA SITRAT PADA PENYIMPANAN SUHU RUANG DAN SUHU REFRIGERATOR DENGAN VARIASI WAKTU PEMERIKSAAN ABSTRAK Latar belakang: Pemeriksaan Activated Partial Thromboplastin Time (APTT) merupakan salah satu uji hemostasis yang digunakan untuk menilai fungsi jalur intrinsik dan jalur bersama pada sistem koagulasi. Tahap pra-analitik, terutama suhu penyimpanan dan penundaan waktu pemeriksaan dapat mempengaruhi stabilitas hasil APTT. Tujuan: Mengetahui perbedaan nilai APTT plasma sitrat pada penyimpanan suhu ruang (20–25 °C) dan suhu refrigerator (2–8 °C) dengan variasi waktu pemeriksaan yaitu ditunda 2 jam, 4 jam, dan 8 jam. Metode: Penelitian Quasi Eksperimental dengan pendekatan repeated-treatment design. Sampel berupa plasma sitrat yang berasal dari 4 mahasiswa sehat dengan teknik simple random sampling. Setiap sampel diberikan 7 perlakuan sehingga diperoleh 28 hasil pemeriksaan APTT. Analisis data menggunakan uji Two-Way ANOVA, dan uji lanjut Post Hoc LSD. Hasil: Rata-rata nilai APTT menunjukkan kecenderungan meningkat seiring penundaan waktu pemeriksaan, terutama pada suhu refrigerator, namun perubahan tersebut tidak bermakna secara statistik. Uji Two-Way ANOVA menunjukkan tidak terdapat perbedaan yang signifikan berdasarkan suhu penyimpanan (p=0,296), waktu pemeriksaan (p=0,239), maupun interaksi keduanya (p=0,909). Kesimpulan: Tidak terdapat perbedaan signifikan nilai APTT plasma sitrat pada penyimpanan suhu ruang dan suhu refrigerator dengan penundaan pemeriksaan hingga 8 jam. Kata kunci: APTT, suhu, penundaan DIFFERENCES IN ACTIVATED PARTIAL THROMBOPLASTIN TIME (APTT) VALUES OF CITRATED PLASMA STORED AT ROOM TEMPERATURE AND REFRIGERATED TEMPERATURE WITH VARIATIONS IN EXAMINATION TIME ABSTRACT Background: Activated Partial Thromboplastin Time (APTT) is a hemostasis test used to evaluate the function of the intrinsic and common coagulation pathways. Pre-analytical factors, particularly storage temperature and delayed examination time, may influence the stability of APTT results. Objective: To determine the differences in APTT values of citrated plasma stored at room temperature (20–25°C) and refrigerator temperature (2–8°C) with examination delays of 2, 4, and 8 hours. Methods: This study employed a quasi-experimental design with a repeated-treatment approach. The samples consisted of citrated plasma collected from 4 healthy students selected through simple random sampling. Each sample received 7 treatment conditions, resulting in 28 APTT examination results. Data were analyzed using Two-Way ANOVA followed by the Post Hoc LSD test. Results: The mean APTT values showed an increasing trend with longer examination delays, particularly at refrigerator temperature; however, the changes were not statistically significant. Two-Way ANOVA demonstrated no significant differences based on storage temperature (p=0.296), examination time (p=0.239), or the interaction between both factors (p=0.909). Conclusion: There was no significant difference in the APTT values of citrated plasma stored at room temperature and refrigerator temperature with examination delays of up to 8 hours.
