Abstract
INHIBITORY TEST OF PAPAYA LEAF EXTRACT (Carica Papaya Linn) AGAINST THE GROWTH OF Shigella dysentriae BACTERIA. Background: Shigella dysenteriae is a pathogenic bacterium that causes infection and has developed resistance to antibiotics. Papaya leaves contain flavonoids, alkaloids, saponins, and tannins, which act as antibacterial agents. Purpose: this study aimed to determine the inhibitory effect of papaya leaf extract on the growth of Shigella dysenteriae. Method: this research is an experimental study using a “Posttest Only Controlled Group Design” with the disk diffusion method at four concentrations: 20%, 40%, 60%, and 80%, chloramphenicol 30 µg was used as the positive control, and 96% ethanol was used as the negative control. Results: the results showed that papaya leaf extract inhibited Shigella dysenteriae, with inhibition zone diameters of 5,88 mm ± 0,10 (20%), 6,28 mm ± 0,10 (40%), 6,68 mm ± 0,10 (60%), and 7,00 mm ± 0,08 (80%). The LSD test showed a significance value of ρ˂α (0.05), indicating a difference in inhibition zones among the various concentrations of papaya leaf extract. Conclusion: of this study is that there are significant differences in the inhibition zones of Shigella dysenteriae growth based on the concentrations of papaya leaf extract. Latar Belakang: Shigella dysentriae merupakan bakteri patogen penyebab infeksi yang mengalami resistensi terhadap antibiotik. Daun pepaya mengandung senyawa flavonoid, alkaloid, saponin, tanin yang berperan sebagai zat antibakteri. Tujuan: penelitian ini bertujuan untuk mengetahui daya hambat ekstrak daun pepaya terhadap pertumbuhan bakteri Shigella dysentrie. Metode: penelitian ini merupakan eksperimen dengan rancangan “Posttest Only Controlled Group Design” menggunakan metode difusi cakram dengan empat konsentrasi yaitu 20, 40, 60, dan 80%, kontrol kerja menggunakan kloramfenikol 30 µg dan kontrol negatif menggunakan etanol 96%. Hasil: penelitian menunjukkan ekstrak daun pepaya mampu menghambat Shigella dysentriae dengan diameter zona hambat 20% sebesar 5,88 mm±0,10, 40% sebesar 6,28 mm±0,10, 60% sebesar 6,68 mm±0,10, 80% sebesar 7,00 mm±0,08. Uji LSD menunjukkan nilai ρ˂α (0,05) sehingga ada perbedaan zona hambat pertumbuhan Shigella dysentriae antara berbagai konsentrasi ekstrak daun pepaya. Simpulan: berdasarkan penelitian ini terdapat perbedaan zona hambat dari variasi ekstrak daun pepaya terhadap pertumbuhan bakteri Shigella dysentriae.
