Abstract

ANTIBACTERIAL ACTIVITY TEST OF ETHANOL EXTRACT OF HYDRANGEA FLOWERS (Hydrangea macrophylla) AGAINST THE GROWTH OF Streptococcus pyogenes ABSTRACT Background: Streptococcus pyogenes is a bacterium that is often the cause of acute respiratory infections. The management that is often used is antibiotics. In addition to antibiotics, natural ingredients can also be used as alternative treatments for infections. Hydrangea flowers contain phytochemical compounds that act as antibacterial substances. Objective: This study aims to determine the antibacterial activity of ethanol extract of hydrangea flowers against the growth of Streptococcus pyogenes bacteria. Methods: True Experimental research with the "Posttest Only Control Group Design" design used disc diffusion method with concentrations of 5, 10, 20, and 40%, positive control using Ampicillin 10 μg and negative control using 96% ethanol each group performed three repeats. Results: The study showed that ethanol extract of Hortensia flowers produced an inhibition zone against Streptococcus pyogenes bacteria with an inhibition zone diameter of 5% of 6.57 mm, 10% of 8.15 mm, 20% of 8.13 mm, 40% of 8.20 mm. Based on the Shapiro Wilk test, the data is normally distributed. The One Way Anova test has a significant difference. The post LSD test p < α (0.05) between positive controls with concentration variations and p > α (0.05) between concentration variations. Conclusion: The results of the study showed that ethanol extract of hydrangea flowers had antibacterial activity in the moderate category, there was a significant difference between positive control and concentration variation while there was no significant difference between variations. UJI AKTIVITAS ANTIBAKTERI EKSTRAK ETANOL BUNGA HORTENSIA (Hydrangea macrophylla) TERHADAP PERTUMBUHAN Streptococcus pyogenes ABSTRAK Latar Belakang: Streptococcus pyogenes merupakan bakteri yang sering menjadi penyebab infeksi saluran pernapasan akut. Penatalaksanaan yang sering digunakan adalah antibiotik. Selain antibiotik, bahan alam juga bisa sebagai pengobatan alternatif untuk infeksi. Bunga hortensia mengandung senyawa fitokimia yang berperan sebagai zat antibakteri. Tujuan: Penelitian ini bertujuan untuk mengetahui aktivitas antibakteri ekstrak etanol bunga hortensia terhadap pertumbuhan bakteri Streptococcus pyogenes. Metode: Penelitian True Experimental dengan rancangan “Posttest Only Control Group Design” menggunakan metode difusi cakram dengan konsentrasi yaitu 5, 10, 20, dan 40%, kontrol positif menggunakan Ampicillin 10 µg dan kontrol negatif menggunakan etanol 96% setiap kelompok melakukan tiga kali pengulangan. Hasil: penelitian menunjukkan ekstrak etanol bunga Hortensia menghasilkan zona hambat terhadap bakteri Streptococcus pyogenes dengan diameter zona hambat 5% sebesar 6.57 mm, 10% sebesar 8.15 mm, 20% sebesar 8.13 mm, 40% sebesar 8.20mm. Berdasarkan uji Shapiro Wilk data berdistribusi normal. Uji One Way Anova terdapat perbedaan yang bermakna. Uji post LSD p < α (0,05) antara kontrol positif dengan variasi konsentrasi dan p > α (0,05) antar variasi konsentrasi. Simpulan: Hasil penelitian menunjukkan ekstrak etanol bunga Hortensia memiliki aktivitas antibakteri dengan kategori sedang, terdapat perbedaan bermakna antara kontrol positif dengan variasi konsentrasi sedangkan antar variasi tidak terdapat perbedaan signifikan. Kata kunci: Uji Antibakteri; ekstrak Hydrangea macrophylla.
