Abstract

COMPARISON OF THE ANTIBACTERIAL ACTIVITY OF GUAVA LEAF EXTRACT (Psidium Guajava L.) USING DISK DIFFUSION AND WELL DIFFUSION METHODS AGAINST Staphylococcus aureus BACTERIA ABSTRACT Background of this study is diarrhea, which remains a global health issue. One of its causes is infection by Staphylococcus aureus, a bacterium that has increasingly developed resistance to antibiotics. This condition highlights the need for alternative treatments, including the use of natural substances such as guava leaves (Psidium guajava L.), which are known to have antibacterial properties. Purpose of this study is comparison of the antibacterial activity of guava leaf extract (Psidium guajava l.) using disk diffusion and well diffusion methods against Staphylococcus aureus bacteria. Research method is a true experimental study with a post-test only control group design. A 25% guava leaf extract was tested in five different volumes: 12.5 µl, 25 µl, 50 µl, 75 µl, and 100 µl. Results showed that the inhibition zone increased with the volume of extract used. The 100 µl volume produced the largest inhibition zones, with an average of 13.21 mm in the disk diffusion method and 19.72 mm in the well diffusion method. A significant difference was observed between the two methods, with the well diffusion method producing larger inhibition zones. Conclusion of this study is that guava leaf (Psidium guajava L.) extract is effective in inhibiting the growth of Staphylococcus aureus, and the well diffusion method is considered more optimal for testing antibacterial activity. DENGAN METODE DIFUSI CAKRAM DAN SUMURAN TERHADAP BAKTERI Staphylococcus aureus ABSTRAK Latar Belakang penelitian ini adalah diare yang masih menjadi masalah kesehatan global, salah satunya disebabkan oleh infeksi bakteri Staphylococcus aureus, yang kini banyak mengalami resistensi terhadap antibiotik. Kondisi ini mendorong perlunya alternatif pengobatan, salah satunya melalui pemanfaatan bahan alam, seperti daun jambu biji (Psidium guajava L.) yang dikenal memiliki kandungan antibakteri. Tujuan penelitian ini untuk membandingkan aktivitas antibakteri ekstrak daun jambu biji (Psidium guajava L.) dengan metode difusi cakram dan sumuran terhadap bakteri Staphylococcus aureus. Metode penelitian ini yaitu eksperimental murni (true experimental) dengan desain post-test only control group design. Ekstrak daun jambu biji (Psidium guajava L.) konsentrasi 25% diuji dalam lima variasi volume, yaitu 12,5 µl, 25 µl, 50 µl, 75 µl, dan 100 µl. Hasil penelitian ini menunjukkan bahwa zona hambat meningkat seiring bertambahnya volume ekstrak. Volume 100 µl menghasilkan zona hambat terbesar dengan rerata, yaitu 13,21 mm pada metode difusi cakram dan 19,72 mm pada metode difusi sumuran. Terdapat perbedaan signifikan antara kedua metode, dengan metode sumuran menghasilkan zona hambat yang lebih besar. Simpulan penelitian ini adalah ekstrak daun jambu biji (Psidium guajava L.) terbukti efektif menghambat pertumbuhan bakteri Staphylococcus aureus, dan metode sumuran dinilai lebih optimal dalam menguji aktivitas antibakteri.
